人参皂甙Rg1和Rb1对脐血CD34+细胞凋亡的影响

【摘要】 目的 探讨人参皂甙Rg1和Rb1对体外培养体系中撤除细胞因子引起的脐血CD34 + 细胞凋亡的干预作用。方法 免疫磁珠法纯化获得脐血CD34 + 细胞,在含有TPO(50μg/L)、SCF(50μg/L)的StemSpan TM SFEM无血清液体培养基中培养7天后,撤除细胞因子实验组加入不同浓度Rg1、Rb1,24h和72h后收集细胞,流式细胞仪FITC—Annexin—V/PI双色荧光标记检测细胞凋亡率,比色法检测Caspase—3活性。结果 撤除细胞因子24h后,撤因子组细胞凋亡率为(24.79±2.80)%,Rgl(2.5mg/L、5mg/L、10mg/L)各组细胞凋亡率均降低,分别为(12.35±2.07)%、(11.87±1.20)%和(13.91±0.50)%(P0.01);Rb1各组(5mg/L、10mg/L)的细胞凋亡率分别为(17.88±0.65)%、(20.61±0.85)%(P0.01);撤除细胞因子24h和72h对照组,Caspase—3活性设为1.00,Rg1—2.5组(0.72±0.09,0.57±0.22,P0.01),Rg1—5组(0.51±0.11,0.42±0.10,P0.01),Rg1—10组(0.63±0.06,P0.01;0.83±0.12,P0.05),Rb1—5组(0.54±0.18,0.32±0.18,P0.01)Rb1—10组(0.87±0.07,P0.05;0.55±0.03,P0.01),Caspase—3活性均低于对照组。结论 人参皂甙Rg1、Rb1能拮抗因撤除细胞因子诱导脐血CD34 + 细胞凋亡,此作用与Caspase—3信号通路有关。

【Abstract】 Objective To investigate the effects of ginsenosides Rg1and Rb1during umbilical cordal blood hematopoietic cells apoptosis induced by growth factors withdrawal.Methods Human umbilical cordal blood CD34 + stem cells were purified and obtained steriley.CD34 + cells were cultured in StemSpan TM SFEM serum—free medium containing cytokines TPO(50μg/L)and SCF(50μg/L)for7days.Then cells were collected and maintained in the same medium with the growth factors deprivation for24h and72h.The experimental groups were respectively treated with Rg1or Rb1with different concentrations.FITC—Annmexin—V/PI flow cyloanalyzer and Caspase—3colorimetˉric assay were respectively used to determine cell apoptosis rate and the activity of Caspase—3.Results 24hours afˉter the cytokines withdrawal,the apoptosis rate in control cells was(24.79±2.80)%;the apoptosis rate in Rg1(2.5 mg/L,5mg/L,10mg/L)groups were(12.35±2.07)%,(11.87±1.20)%and(13.91±0.50)%(P0.01);in Rb1groups(5mg/L,10mg/L)were(17.88±0.65)%,(20.61±0.85%)(P0.01)respectively.24hours and72hours after cytokines deprivation,Caspase—3activities in control groups were taken as1.00,the values in Rg1—2.5group were0.72±0.09and0.57±0.22(P0.01),in Rg1—5group were0.51±0.11and0.42±0.10(P0.01),in Rg1—10group were0.63±0.06(P0.01)and0.83±0.12(P0.05);in Rb1—5group were0.54±0.18and0.32±0.18(P0.01),in Rbl—10group were0.87±0.07(P0.05),0.55±0.03(P0.01).Conclusion These results demonstrated that apoptosis induced by growth factors withdrawal of umbilical cordal blood hematopoietic cells can be inhibited by Rg1and Rb1and this effect was involved with Caspase—3signal pathway.

细胞凋亡细胞在一定的生理或病理条件下,遵循自身的程序的一种主动性的死亡过程,它涉及细胞内一系列的信号转导和调控过程。在造血过程中,细胞凋亡在调控细胞增殖、分化的平衡中起重要作用。人参“补气养血”的功效虽然已经在临床上应用已久,但机理并未阐明。近年来对人参的主要成分人参皂甙的研究很多,多集中在促进 ˇ 基金项目:天津市科委自然科学基金资助项目(编号:013608511)造血细胞增殖,诱导髓性白血病细胞凋亡,增强机体免疫功能等方面。国内学者研究发现人参皂甙不但能促进造血干/祖细胞增殖,并且能诱导定向分化 [1] 。但人参皂甙Rg1和Rb1对造血细胞凋亡的影响尚未见报道。本研究观察了人参皂甙Rg1和Rb1对撤除细胞因子引起的脐血造血细胞凋亡的拮抗作用,以期在细胞水平上为人参皂甙“补气养血”功效的作用机制提供更多的实验室证据。
