蝙蝠葛果提取物对小鼠S180荷瘤的抑制作用及耐缺氧作用

【摘要】 [目的]观察蝙蝠葛果提取物对移植性小鼠S180荷瘤肿瘤生长及耐缺氧的影响.[方法]建立移植性小鼠S180肉瘤模型,随机分为对照组环磷酰胺组、蝙蝠葛果提取物大、小剂量(500,250mg/kg)组.将不同剂量蝙蝠葛果提取物灌胃给予荷瘤小鼠10d后,停药1d,次日处死,计算肿瘤抑制率,称取胸腺及脾脏质量;检测灌胃给予蝙蝠葛果提取物小鼠的耐缺氧存活时间.[结果]蝙蝠葛果提取物剂量小鼠S180荷瘤质量明显减轻,肿瘤抑制率为40.7%,并且胸腺质量显著高于对照组蝙蝠葛果提取物大、小剂量小鼠缺氧存活时间均明显延长.[结论]蝙蝠葛果提取物对S180荷瘤小鼠具有抑制肿瘤的生长及耐缺氧作用.

ABSTRACT:OBJECTIVETo study the functions of tumor inhibition and toleration anoxia of the extract of Menispermum Dahuricum fruits (MDF) for the transplanted S180 bearing cancer in mice.METHODSS180 sarcoma transplanted models in mice were established and randomly divided into the model group, cyclophosphamide group and large, small dose of extraction (500, 250mg/kg) of MDF groups. The different concentration of MDF extraction was administrated by oral to S180 bearing cancer in mice for 10 day and stoped administration for 1 day, and then the mice was sacrificed next day to calculate rate of inhibiting tumor growth and ratio of thymus gland and spleen and examine the survival time of toleration anoxia.RESULTSThe weight of S180 bearing cancer in mice was obviously reduced in group of large dose of MDF extraction, the rate of inhibiting tumor growth was 40.7%, the index of thymus gland was higher in MDF extraction group than in the model control group, and the survival time of toleration anoxia extended in groups of MDF extraction.CONCLUSIONThe extraction of MDF has the function of inhibiting tumor growth and toleration anoxia in the S180 bearing cancer in mice.

蝙蝠葛( Menispermum dahuricum DC.)是防己科植物,又名北斗根、蝙蝠藤、金丝钓葫芦、黄条香、防己葛、黄根、野鸡豆子、狗屎豆、马串铃等.蝙蝠葛味苦,性寒,无毒,有祛风清热,消肿止痛作用,可用于治疗扁桃体炎、咽喉炎、风湿痹痛、麻木、水肿、脚气、肠炎及胃痛腹胀等[1].蝙蝠葛有抗心率失常、降血压、抗血栓、抑制血小板聚集及抗炎镇痛等作用.国外多项研究表明,蝙蝠葛根、茎提取物对耐缺氧和抗肿瘤有较好的疗效[2].本研究建立了移植性小鼠S180肉瘤模型,探讨了蝙蝠葛果提取物肿瘤生长及耐缺氧的影响.

1 材料与方法。

1.1 材料 蝙蝠葛果提取物由延边大学药学院植物化学实验室制备并提供;环磷酰胺粉针剂为山西普德药业有限公司产品,加入适量生理盐水配制成1.25g/L,备用.实验动物取昆明种小鼠,雄性,体重为18~22g,由延边大学医学部实验动物科提供.S180腹水荷瘤模型由延边大学基础医学院肿瘤研究中心制作并提供.GB204分析天平为瑞士梅特勒托利多仪器(上海)有限公司产品.

1.2 方法。

1.2.1 肿瘤抑制率的测定 将S180荷瘤小鼠腹腔肿瘤细胞接种于小鼠腹腔连续传代,无菌抽取7d龄小鼠腹腔肿瘤细胞,加入生理盐水调整细胞中肿瘤细胞至4×106个/mL,以1∶3的比例混匀,向每只小鼠右腋下注射0.2mL肿瘤细胞悬液[3],制作移植性小鼠S180荷瘤模型.将模型小鼠随机分为4个组,即对照组环磷酰胺组、蝙蝠葛果提取物大、小剂量(500,250mg/kg)组,每组各为10只;各组小鼠体重间无显著性差异.对照组小鼠灌胃给予生理盐水,环磷酰胺(1.25g/L)组小鼠腹腔注射给予环磷酰胺蝙蝠葛果提取物大、小剂量灌胃给予相应剂量蝙蝠葛果提取物;各组小鼠灌胃剂量均为每日20mL/kg.连续给药10d,第11日停药,次日脱颈处死小鼠,剥下肿瘤,称其质量,计算肿瘤抑制率.

肿瘤抑制率(%)=对照组平均肿瘤质量—药物组平均肿瘤质量对照组平均肿瘤质量×100%。
