开胃进食汤超微配方颗粒对正常小鼠胃肠动力的影响

【摘要】; 目的 研究开胃进食超微配方颗粒对正常小鼠胃排空、小肠推进活动的影响。方法 采用开胃进食汤传统汤剂和健胃消食片对照,观察开胃进食超微配方的等倍、1/2倍和1/4倍不同剂量小鼠胃内甲基橙残留率和小肠质量、小肠碳末推进率的影响。结果 开胃进食超微配方颗粒各组胃内甲基橙残留率较正常组低(P<0.05,P<0.01),等倍和1/2倍剂量小肠碳末推进率较正常组高(P<0.05,P<0.01),1/2倍剂量小肠质量较正常组高(P<0.05),而开胃进食汤传统汤剂组与正常组比较,以上指标无差异。结论 开胃进食超微配方颗粒对正常小鼠具有促进胃肠动力作用,且以1/2倍剂量为优;超微配方颗粒制剂可能与诸相关药物如藿香、木香、丁香等药物挥发油成分作用的关系,从而提高了其传统方剂的药理作用。

; 〔Abstract〕 Objective To study the effects of Kaiwei Jinshi decoction with the ultrafine particle on the normal gastric evacuatian and small intestinal propulsion in mice. Methods The untrafine and traditional decoctions were compared, the methyl orange residual rate in stomach, small intestine weight and small intestine carbon powder propulsion rate were measured with the isodose, 1/2 and 1/4 dosages of both ultrafine particle and traditional type. Results The methyl orange residual rate in stomach in all groups of ultrafine particle were lower than that in normal group(P<0.05 or P<0.01), the carbon powder propulsion rate in small intestine with same and 1/2 dosages were higher than that in normal group (P<0.05 or P<0.01), the weight of small intestine with 1/2 dosages was more than that in normal group (P<0.05), while, there was no difference between the traditional herbal medicine and ultrafine particle groups. Conclusion The ultrafine decoction can improve the gastrointestinal power, especially with 1/2 dosages; the ultrafine particle preparation enhances the pharmacological action owing to possibly retaining the component of volatile oil such as Clove, Aucklandiae and Agastache rugosa.。

; 开胃进食汤系《医宗金鉴》治疗脾胃虚弱的有效方剂。近十年的文献资料,有关开胃进食汤的报道仅限小样本临床观察和个案病例报道,我们拟用其超微配方颗粒与传统汤剂比较,从对胃肠动力的调节作用初步研究其药理作用。现将实验方法及结果报道如下。

; 1 材料与方法。

; 1.1; 材料。

; 1.1.1; 动物; NIH小鼠120只,体质量18~22 g,雌雄各半。购自湖南农业大学实验动物中心,合格证:SCK(湘)2003-003。

; 1.1.2; 药物; 开胃进食汤(药物组成:白参12 g,白术10 g,茯苓12 g,陈皮9 g,半夏6 g,丁香6 g,木香6 g,藿香10 g,莲子15 g,厚朴9 g,砂仁6 g,麦芽12 g,神曲10 g,甘草10 g)传统汤剂超微配方颗粒均由湖南中医药研究院超微中心提供,其中传统汤剂每毫升生药0.93 g,药物浓度按《中药药理研究方法学》人与小鼠药物剂量换算表,折算成每毫升生药0.62 g(以下换算标准同);超微配方颗粒每克含生药4.5 g,药物浓度分别为等倍剂量每毫升生药0.6 g,1/2倍剂量每毫升生药0.3 g,1/4倍剂量每毫升生药0.15 g;健胃消食片,每片含生药0.8 g,江西江中制药厂出品,批号:0612016,药物浓度为每毫升生药 0.037 6 g。

; 1.1.3; 试剂与仪器; 0.1%甲基橙溶液、30%活性炭混悬液,天津市大茂化学试剂厂生产;721分光光度计,上海第三分析仪器厂生产。

; 1.2; 方法。

; 1.2.1; 动物分组及给药方法; 胃排空及小肠推进实验,各取NIH小鼠60只,体质量18~22 g,雌雄各半。运用随机数字表随机分为6组,每组10只,分别为空白组、健胃消食片组、开胃进食汤传统汤剂组(简称传统汤剂组)、开胃进食配方颗粒高、中、低剂量组,分别相当于人体的等倍、1/2倍、1/4倍药物剂量(简称超微等倍组、超微1/2倍组、超微1/4倍组),连续灌胃给药5 d(0.25 mL/10 g),空白对照组给予等容积生理盐水。
