不同抗原致敏树突状细胞对荷瘤大鼠经射频消融后免疫机能的影响

【摘要】 目的 本研究在大鼠Walker 256实体瘤射频消融术(RFA)后应用不同肿瘤抗原致敏的DC瘤苗,探讨其对大鼠肿瘤免疫的不同影响。方法 38只荷Walker 256实体瘤SD大鼠随机分成3组:对照组行RFA治疗实验组1行RFA+冻融抗原致敏DC治疗实验组2行RFA+热休克冻融抗原致敏DC治疗治疗前及治疗后第4天分别测量各组大鼠外周血中CD4+ T淋巴细胞、CD8+ T淋巴细胞及CD4+ /CD8+比值,记录大鼠荷瘤生存期。结果 治疗后各组荷瘤大鼠外周血中CD8+T细胞均呈下降改变,其中,下降的程度为实验组2实验组1对照组(P0.05);CD4+/CD8+比值的变化,实验组2的升高与其他两组相比差异有统计学意义 (P0.05);实验组1的升高与对照组比差异无统计学意义(P0.05)。荷瘤生存期:实验组2实验组1对照组(P0.05)。 结论 在RFA后应用不同抗原致敏的DC可不同地程度改善大鼠的抗肿瘤免疫,冻融休克抗原致敏DC较未热休克冻融抗原致敏DC能更有效地激发荷瘤大鼠的抗肿瘤免疫机能。   【关键词】树突状细胞;射频消融;免疫      【Abstract】 Objective Dendritic cell (DC) lose the nomal function in tumor host. The tumor—antigen pulsed DCs obtained in vitro can effectively improve the immulogical state. The purpose of this study is to invest the immunological influence of different pulsed dendritic cells (DCs) on rats after radiofrequency ablation (RFA) of the tumor.Methods 38 rats with Walker 256 solid tumor were divided into four groups: control groupwas treated with RFA only; experimental group 1was treated with un—heat—shocked freeze—thraw antigen pulsed DCs after RFA; experimental group 2was treated with heat—shocked freeze—thraw antigen pulsed DCs after RFA. Before treatment and 4 days after treatment, the CD4+ T cell and CD8+ T cell were checked with cytometric analysis, and CD4+/CD8+ were evaluated. The suvival duration of the rats were recorded. Results After treatment, For the degree of the decrease of CD8+ T cell, there were significant difference between the different group with the degree of group 2 group 1 control (P0.05). The survival duration of the rats in group 2 is the longest and then that of group 1 (P0.05). Conclusion Using different antigen—pulsed DCs can improve the antitumor immunological function of the rats after RFA. Heat—shocked freeze—thraw antigen pulsed DCs can more effectively do that than un—heat—shocked freeze—thraw antigen pulsed DCs.   【Key words】Dendritic cell; Radiofrequency ablation; Immunology      在肿瘤治疗当中进一步激发、增强机体对肿瘤的主动免疫反应,有可能在延缓肿瘤生长、杀灭体内微小转移灶或残留癌细胞、防止肿瘤复发和转移等方面起到重要的作用[1—2]。树突状细胞(DCs)是目前发现的功能最强的抗原提呈细胞,具有强大的激活初始型T细胞的能力,是目前肿瘤免疫治疗的研究热点之一。本研究在荷瘤大鼠实体瘤经射频消融(RFA)治疗后应用不同抗原致敏的DC瘤苗,旨在探讨不同的DC瘤苗对荷瘤大鼠肿瘤免疫机能的影响。   1 资料与方法   1.1 实验动物及实体瘤模型的制备   1.1.1 实验动物 SD大鼠38只,体质量80~100 g,雌雄不限,购自广州中医药大学实验动物中心。   1.1.2 实体瘤模型的制备 将Walker 256细胞(购自第二军医大学长海医院)悬液注入大鼠腹腔,约7 d后获得大鼠Wakler 256腹水型肿瘤模型;抽取约1 ml 腹水(血性为佳)注入大鼠右侧腋窝皮下,5 d后获得大鼠Walker 256实体瘤模型。   1.1.3 实验分组 将成瘤大鼠随机分成3组: 对照组(n =10),行RFA治疗实验组1(n =10),行RFA+冻融抗原致敏DC治疗实验组2(n=18),行RFA+热休克冻融抗原致敏DC治疗。   1.2 DC瘤苗的制备   1.2.1 DC的培养及鉴定 断颈处死大鼠后,以75 %酒精浸泡10 mim,取大鼠后腿股骨及胫骨,PBS液冲洗骨髓腔,经离心及红细胞裂解液等处理分离获骨髓单核细胞,培养液加入细胞因子rGM—CSF 5 ng/ml和IL—4 5 ng/ml(R&D 公司),置于培养箱中培养,隔天进行半量换液。收集培养8 d的DC行Giemsa’s染色光镜下观察;同时收集DC进行流式细胞免疫学检测表面特异性抗原OX6、OX62和细胞因子CD86、CD80。   1.2.2 DC瘤苗的制备和检验 Walker 256细胞株离心后42 ℃温浴1 h(热休克),反复冻融,或未经热休克处理的Walker 256 细胞反复冻融后,离心,取上清,-80℃冰箱保存备用。DC培养第7 天加入预先制备的肿瘤抗原,置于培养箱中培养,隔天收集。体外进行混合淋巴细胞反应试验和MTT法淋巴细胞杀伤试验。。
