沙棘汁对铅所致小鼠脑组织生物化学指标的影响

[摘要] [目的] 研究沙棘汁对醋酸铅所致小鼠脂质过氧化胆碱酯酶和单胺类神经递质含量改变的影响. [方法] 制作铅中毒小鼠模型,灌胃给予不同剂量的沙棘汁,利用硫代巴比妥酸比色法、碱性羟胺法及荧光法分别测定小鼠脑组织过氧化脂质胆碱酯酶及单胺类神经递质含量. [结果] 铅组小鼠脑组织内MDA含量明显升高,200,400g/L沙棘汁降低铅致MDA含量升高,且呈剂量—效应关系;铅组小鼠脑组织胆碱酯酶活性明显升高,单胺类神经递质中除DA外NE,5—HT,5—HIAA含量均明显下降,400g/L沙棘汁抑制铅致脑组织内多种神经递质含量的变化. [结论] 沙棘汁可拮抗铅所致小鼠脑组织脂质过氧化胆碱酯酶和单胺类神经递质含量的改变.

Effects of the Hippophae Rhamnoides L juice on lead—induced change of lipid peroxidation,acetylcholinesterase and catecholamine neurotrans—mitters in mice brain。

ABSTRACT:OBJECTIVE To study the effects of the Hippophae Rhamnoides L juice on lead—induced change of lipid peroxidation,acetylcholinesterase(AchE)and catecholamine neurotransmitters in mice brain.METHODS Mice were administrated peritoneal injection with lead acetate,then mice were given various dosages of Hippophae Rhamnoides L juice by gavage.The value of MDA was determined by the thiobarituric acid colorimetricmethod,and the activity of AchE was determined by the alkaline hydroxylamine method,and contents of NE,DA,5—HT were determined by the flourimetric method.RESULTS The MDA levels were increased in lead—treated mice,which were reduced by200,400g/L Hippophae Rhamnoides L in dose—dependent manner.The activities of AchE was significantly increased in the lead—treated group,which were decreased by400g/L Hippophae Rhamnoides L.The levels of NE,5—HT,5—HIAA were significantly decreased in the lead—treated mice,and this decreases were antagonized by400g/L Hippophae Rhamnoides L,except for that in dopamine.CONCLUSION Hippophae Rhamnoides L juice has protective effects against the lead acetate—induced changes in neurobiochemical parameters.

铅是广泛存在于自然环境中的强神经毒物,可影响神经系统的许多功能.动物实验及临床观察结果表明,铅中毒可引起行为和智力障碍,可改变一些酶的活性,减少突触前神经递质的释放,影响自由基代谢 [1,2] .沙棘为胡颓子科酸刺属灌木,含有多种维生素(E,C,B 1 ,B 2 ,B 6 )、氨基酸、黄酮、脂肪油及微量元素等物质,具有降血脂、抗心律失常、抗氧化、抗癌、抗过敏及增强免疫功能等功效 [3] .本实验选用神经生物化学实验方法,观察了沙棘对铅致小鼠脂质过氧化胆碱酯酶和单胺类神经递质含量改变的影响.

1 材料与方法。

1.1 材料。

1.1.1 试剂 沙棘汁为采集于吉林省珲春市郊产沙棘果经压榨及过滤而得原汁,加水配制成200,400g/L;醋酸铅为哈尔滨市化工试剂厂产品,临用前用双蒸水配制成浓度为10mmol/L的溶液,以每g体重10μL的剂量行腹腔注射;NE,DA,5—HT,5—HIAA购自Sigma公司;MDA测试盒、胆碱酯酶测试盒、蛋白质定量试剂盒购自南京建成生物工程公司.

1.1.2 实验动物 取昆明种小鼠,体重为20~22g,雌雄兼用,由延边大学医学部实验动物科提供.

1.2 方法。

1.2.1 过氧化脂质含量的测定 取昆明种小鼠50只,随机分为5组,每组各为10只.1)对照组:每日腹腔注射等体积生理盐水10mL/kg;2)铅组:每日腹腔注射醋酸铅溶液10mg/kg;3)400g/L沙棘汁组:每日灌胃给予400g/L沙棘汁10mL/kg;4)铅加200g/L沙棘汁组:上午灌胃给予200g/L沙棘汁10mL/kg,下午腹腔注射醋酸铅溶液10mL/kg;5)铅加400g/L沙棘汁组:上午灌胃给予 400g/L沙棘汁10mL/kg,下午腹腔注射醋酸铅溶液10mL/kg.连续给药20d,其后对相应组别小鼠隔日分别给予200及400g/L沙棘灌胃,给予对照组小鼠等量生理盐水灌胃,共5次.实验第31日处死动物,取脑,称其质量,在预冷的0.32mol/L蔗糖,10mmol/L PBS(pH值为7.4)中以1∶10的比例匀浆,以3500r/min离心10min,取上清液,用MDA测试盒测定过氧化脂质含量.

1.2.2 胆碱酯酶活性的测定 分组、染毒方式及给药方法同1.2.1项下方法.实验第31日处死动物,取脑,称其质量,在70mmol/L PBS中匀浆,离心30min,取上清液50μL置于反应管内,利用胆碱酯酶测试盒测定胆碱酯酶活性 [4] .
